      对海南岛西线海岸采集的红树林植物样品共附生放线菌进行分离、鉴定及抗菌活性分析。采用4种分离培养基分离10份红树林植物茎的共附生放线菌,利用16S rRNA序列分析对分离的放线菌进行鉴定,采用滤纸片法扩散法对分离放线菌的发酵产物进行抗菌活性分析。结果共分离获得63株放线菌,初步确定16株代表性放线菌的分类地位。其中,13株放线菌属于链霉菌属,2株放线菌属于小单孢菌属,4株链霉菌为潜在的新物种。抗菌活性测试显示16株代表性放线菌的发酵产物对至少一种指示菌具有抗菌活性。从菌株HNM0645的发酵粗提物中分离获得了一个化合物A,经过结构鉴定为已知化合物K-252a。通过测试不同浓度化合物A对山药炭疽病原菌的抑菌率,利用Excel求出毒力回归方程,计算出EC50值为286.61μg/mL,化合物A对供试细菌没有明显的抑菌活性。
英文摘要:
      The isolation, identification and antibacterial activity of actinomycetes associated with mangrove plant samples collected from the west coast of Hainan were conducted.The actinomycetes were isolated from 10 mangrove plant samples by using four isolation media. The isolated actinomycetes were identified by 16S rRNA sequence analysis, and Antibacterial activity of fermentation products of the actinomycetes were analyzed by filter paper diffusion method. The results showed that A total of 63 actinomycetes were obtained from 10 mangrove plant samples. The taxonomic status of 16 representive actinomycetes were determined. Among them, 13 isolates belong to the genus Streptomyces, 3 isolates belong to the genus Micromonospora and 4 Streptomyces isolates represent potential novel species.The antibacterial activity test showed that the fermentation products of 16 representive actinomycetes have antibacterial activity against at least one indicator bacteria. The compound A was isolated from the crude fermentation extract of strain HNM0645, which was identified as known compound K-252a by structure. Through testing the inhibitory rate of compound A against yam anthracnose pathogens, using Excel to calculate the virulence regression equation, the EC50 value was calculated to be 286.61μg / mL. Compound A had no significant antibacterial activity against the tested bacteria.
