      为探索新型大环内酯类杀虫剂天维菌素对蚊虫防控的应用前景,进一步了解天维菌素对蚊虫的作用机制,本文利用转录组测序技术研究埃及伊蚊幼虫参与药物代谢及靶标等生物学过程的基因。利用高通量测序获取给药前后埃及伊蚊的差异表达基因,进一步进行Gene Ontology(GO)及KEGG富集分析,并利用Real-time Quantitative PCR(rt-qPCR)验证目标基因表达情况。结果显示,与对照组相比,天维菌素处理后埃及伊蚊幼虫中有2647个基因差异显著表达,其中上调基因有697个,下调表达的基因有1950个。 Gene Ontology分析显示,测序基因主要注释到细胞过程、代谢过程、膜、结合和催化活性等功能类。KEGG通路主要富集在药物代谢、免疫应答、生物合成以及消化与吸收等过程。选取GST-1、EST2等六个目标基因进行rt-qPCR验证,表达水平与测序结果一致。本实验通过转录组测序技术初步探究埃及伊蚊幼虫参与天维菌素胁迫的基因集,为进一步了解天维菌素的潜在靶点及抗药性基因奠定了基础。

      In order to investigate the gene expression differences of Aedes aegypti at the transcriptome level before and after administration of tenvermectin, RNA-Seq was used to sequence the transcriptome of Aedes aegypti larvae before and after the treatment with the drug. The gene expression differences of Aedes aegypti before and after the treatment were then analyzed, and verified by Real-time Quantitative PCR. The results showed that, compared with the control group, there were 2647 genes with significant difference in expression in the larvae of Aedes aegypti treated with tenvermectin, of which 697 were up-regulated genes and 1950 were down-regulated genes. Six genes were randomly selected and verified by Real-time Quantitative PCR. The results were consistent with RNA Seq results. Gene ontology analysis showed that sequencing genes were annotated into the following groups: cellular process, metabolic process, membrane, binding and catalytic activity. The differential gene KEGG Pathway is mainly enriched in drug metabolism, immune response, biosynthesis, digestion and absorption. In this study, RNA Seq was used to screen out the genes of different expression of Aedes aegypti before and after treatment with tenvermectin, which privides insight into understanding the toxicological mechanism of tenvermectin in Aedes aegypti and finding the drug resistance related genes of Aedes aegypti..
